This product is a long-acting tricyclic antihistamine, which competitively inhibits histamine H 1 receptor and inhibits allergic symptoms caused by histamine. This product has no obvious anticholinergic and central inhibitory effects.

pharmacokinetics
Oral absorption is rapid on an empty stomach. It takes effect in1~ 3 hours, reaches the maximum effect in 8 ~12 hours, and lasts for more than 24 hours. Food can delay the drug peak time and increase AUC. In normal adults, the T 1/2 of loratadine is 28 (8.8 ~ 92) hours.
In patients with chronic alcoholic liver disease, the AUC and peak concentration of loratadine are twice as high as those of normal people, and the T 1/2 of loratadine and its metabolites are 24 hours and 37 hours respectively, which can be prolonged with the severity of liver disease.
The protein binding rates of loratadine and its metabolites are 97% and 73% ~ 77%, respectively.
This product and its metabolites are not easy to penetrate the blood-brain barrier, and mainly act on peripheral H 1 receptor sites.

Co-administration with macrolide antibiotics, cimetidine, theophylline and other drugs can also inhibit the metabolism of loratadine.
overdose
In case of overdose poisoning, if the patient is awake, he can induce vomiting. Normal saline can be used for gastric lavage, and activated carbon can be used to adsorb drugs. Salt laxatives (sodium sulfate) can also be considered to prevent drugs from being absorbed in the intestine. Hemodialysis cannot eliminate this product, and it is not clear whether peritoneal dialysis can eliminate this product.
