How to prepare hydrolyzed casein (MH) agar medium?
Formula milk powder (per liter):

Agar 17g

300 grams of beef (extracted from it)

Soluble starch1.5g.

Casein hydrolysate17.5g

The final ph value is ph 7.3±0.2.

How to use:

1. Weigh 38g of this product, add distilled water or deionized water 1 L, stir, heat and boil until it is completely dissolved, package in triangular bottles, and autoclave at 12 1℃15min.

2. Preparation of test bacteria solution: 3-5 colonies with similar morphology were selected from TSA plate and inoculated in MHB or TSB medium. After culturing at 3665438 0℃ for 4-6 hours, the turbidity reached 0.5 Maxwell units.

3. Inoculation: soak the bacterial solution with good turbidity with sterile cotton swabs, gently rotate the pressure in the tube wall to remove excess bacterial solution, and then gently smear the culture medium, three times per plate. After each smear, the Petri dish should be rotated 60 degrees before the next smear. Each quality control strain was coated with three plates.

4. Patch: Leave the inoculated dish at room temperature for a period of time, and after it is slightly dry, use sterile tweezers to evenly paste antibiotic paper on the agar surface of the dish. The center distance between the pieces of paper should not be less than 24 mm, and the distance between the pieces of paper and the edge of the plate should not be less than 15 mm. Once the paper touches the agar surface, it can't move any more, so the drug has begun to spread.

5. Culture: After pasting the paper, turn the plate upside down within 15-30 minutes, culture at 36 1℃ for 16-24 hours, and observe the results. Plate culture should not be stacked, but two plates should overlap.
