Glycosylation will increase the molecular weight of protein. In SDS-PAGE, protein is usually separated according to their molecular weight. Because the added sugar chain increases the overall molecular weight of protein, the migration of glycosylated protein on the gel may be inconsistent with the expected molecular weight based on protein amino acid chain.
2. Influence 2: Charging state
Although the existence of SDS can usually make protein have a uniform negative charge, so that the electrophoresis process is mainly based on the difference of molecular weight, in some cases, highly glycosylated protein may show unexpected migration behavior, which may be due to the influence of charge masking or addition caused by glycosylation.
