Method for cultivate Cordyceps sinensis
1. Take fresh Cordyceps sclerotium, disinfect the surface under aseptic conditions, then wash it with sterile water, then cut it into small pieces with a scalpel, and then inoculate it on the seed slant culture medium. The culture medium consists of peptone 10g, glucose 40g, potassium dihydrogen phosphate 1g, magnesium sulfate 0.5g, egg yolk 1g and vitamin B65438+. At the temperature of 24 ~ 26℃, about 15d can emit cordyceps similar to Penicillium.
2. Moth larvae like to eat sorghum and Polygonum viviparum rich in starch. Firstly, moth larvae are fed with this feed in large quantities, and the larvae pupate into adults at 28 ~ 30℃. Spraying Cordyceps before pupation, the infected larvae can continue to grow Cordyceps after death.
The formula of culture medium in shake flask and fermentor is corn flour 2% (boiled for 30 minutes, filtered with gauze to get juice), sucrose 2%, peptone 1%, yeast powder 0.5%, potassium dihydrogen phosphate 0. 1% and magnesium sulfate 0.05%.
Shake flask culture adopts 500ml triangular flask culture medium 100 ~ 150 ml, inoculates slant strain, shakes flask culture at 150r/min, and oscillates at 25℃ for 4 days, and can continue to expand the culture to the required liquid volume with 1000ml or 5000ml triangular flask, and then transfer to fermentor.
3. Use a 500L fermentor with 300L feeding, inoculation amount 10%, temperature 24 ~ 26℃, tank pressure 29.4 ~ 49.1kpa (0.3 ~ 0.5 kg/cm2), stirring speed 180 r/min. The fermentation broth of Cordyceps sinensis was placed in a concentration tank and concentrated in vacuum at 77.14 ~ 79.5 kPa (580 ~ 600 mmHg), with evaporation of 300kg/h and temperature of 60 ~ 62℃.
