How to preserve Volvariella volvacea strains?
Volvariella volvacea is one of the edible fungi with the highest demand temperature. Volvariella volvacea strains are not tolerant to low temperature, and will lose their vitality or even die if they are stored below 10℃ for a long time. Therefore, it can't be stored in the refrigerator at 0 ~ 4℃ like other strains, and many strain producers have not mastered this technology. The preservation methods of Volvariella volvacea strains include subculture preservation, liquid paraffin covering preservation, cultivation preservation, freeze-drying liquid nitrogen ultra-low temperature preservation, suspension preservation, natural matrix simple preservation and so on. The most commonly used preservation method in production is subculture preservation. According to the different components of the culture medium, there are two ways of subculture and preservation:

(1) tube slant inoculation culture preservation

Inoculate the Volvariella volvacea strains to the PDA test tube slope, culture at 30℃, and store them at about 20℃ after the mycelium is full of culture medium, at least not lower than 15℃. In order to reduce the loss of water in the substrate during preservation, the strains are usually placed in plastic bags, or the nozzle can be sealed with rubber plugs, and the tubes are rotated every 2 ~ 3 months 1 time.

(2) slant inoculation, culture and preservation of rice straw

95% of rice straw powder, 2% of gypsum, and 3% of bran or rice bran. The pH is adjusted to 8-9 with lime, and the water content is 55%-60%. Put the prepared culture medium into a test tube with a diameter of 25 mm and a length of 250 mm, and the filling amount is 2/3 of the length of the test tube, and seal the nozzle with a cotton plug. After sterilization, the strains of Volvariella volvacea were inoculated and cultured at 33℃. When the hyphae were full of test tubes, rubber plugs were used instead of cotton plugs to seal the nozzle. Store at 1 5 ~ 20℃, and change tubes every 3 ~ 6 months1time.
