In order to ensure that there is enough oxygen in the bacterial culture room, the room must be ventilated twice a day for 20 to 30 minutes each time. The inoculated material bag is called mycelium or bacteria bag. It is the material basis for the growth of fruiting bodies. The quality of mycelium cultivation is directly related to the yield and quality of black fungus. The cultivation period of the bacteria bag takes 40 to 60 days. This period is called the bacteria development and bacteria cultivation stage. The specific methods are as follows:

(1) The bacteria bag discharge culture room should be pre-disinfected. The method can refer to inoculation and disinfection. requirements. After the fungus bags enter the room, long bags should be discharged lying down, or discharged in a "well" shape overlapping each other. Short bags and bottles should be discharged vertically. For inoculation in early spring, due to the low indoor temperature, the bags can be stacked and stacked on 1 to 2 racks, covered with a film, and allowed to grow for 2 to 3 days to increase the bag temperature and accelerate mycelial colonization, and then discharge them separately. (2) Adjusting the temperature requirements for the growth of thermophilic black fungus mycelium should be controlled according to different growth stages.

①Germination period: Within 15 days after inoculation, the indoor temperature should be 26 to 28°C for the first 3 days, so that the mycelium can eat food faster, colonize and spread, and occupy the culture material in the most suitable environment. It creates advantages and reduces bacterial contamination, but it should not exceed 30℃. From the fourth day to the 15th day, as the mycelium grows and develops, the temperature inside the bag gradually rises. Generally, the bag temperature will be 2°C higher than the room temperature, so the room temperature should be adjusted to 25-26°C.

② Robust period: From the 16th to the 35th day, it is the stage with the strongest ability of mycelium to decompose and absorb nutrients. The mycelium is stretched, strong and strong, the metabolism is accelerated, the bag temperature continues to rise, and the room temperature is 23 ~24℃ is appropriate.

③Maturation period: After 35 days, the hyphae enter the physiological maturity stage, which is about to transition from vegetative growth to reproductive growth. The appropriate room temperature is 18~20℃. To control the temperature well, you must hang a thermometer indoors and observe temperature changes at any time. Methods for adjusting temperature: When air conditioning is not widely used in rural areas, the main method is to rely on the switches of doors and windows. In mountainous areas, the temperature of inoculated bacteria in early spring is low, so you can close the door or use charcoal fire to heat it up. If coal is burned, an exhaust pipe should be installed to prevent carbon dioxide from harming the mycelium. When closing the door to increase the temperature, be sure to pay attention to ventilation. At this time, the temperature drops, but it can still rise after closing the door. (3) Humidity control The mycelium cultivation stage requires indoor dryness. The relative humidity of the air should be between 55% and 65%, and should not exceed 75% in the later period. The humidity is high for two days. In addition to opening the doors and windows at any time, increasing the ventilation volume and frequency, you can also sprinkle some lime powder on the floor of the culture room to reduce the relative humidity of the air. (4) It is advisable to stay dark and avoid light. The mycelium cultivation stage requires a dark environment, because as soon as the black fungus mycelium comes into contact with light, it is easy to form ear buds. If the hyphae appear ear buds before physiological maturity, the yield will be affected. For this reason, doors and windows should be covered with black cloth, but good ventilation should be maintained. When the mycelium grows to 85% to 90% away from the bottom of the bag and bottle, it can be exposed 2 days in advance to induce the formation of ear buds. (5) Ventilation and oxygenation. Black fungus is an aerobic fungus. The entire growth and development process requires fresh air to ensure sufficient oxygen to maintain normal metabolism. For this reason, attention must be paid to ventilation 1 to 2 times a day for 30 minutes each time to promote mycelial growth.
