Beef paste 0.3g, peptone 1.0g, sodium chloride 0.5g, agar 1.5g, water 100ml.
Add water 100 ml into the beaker, add beef paste, peptone and sodium chloride, mark the beaker with crayon, and heat it on the fire. After the components in the beaker are dissolved, add agar and stir constantly to avoid sticking to the bottom. After the agar is completely dissolved, make up the water loss, adjust the pH value to 7.2 ~ 7.6 with 10% hydrochloric acid or 10% sodium hydroxide, pack them in each test tube, add cotton plugs, and sterilize with high-pressure steam:1.05kg per square centimeter,12/kloc-.
2. Bovine heart culture medium
Take 250g of fresh beef heart (excluding fat and blood vessels), finely chop it into minced meat with a knife, and then add 500ml of distilled water and 5g of peptone. Mark the culture medium beaker, boil it, and simmer for 2 hours. Filtering, drying the filtered minced meat, and adjusting the pH value of the filtrate to about 7.5. Add 10 ml broth and a small amount of minced dried beef heart into each test tube, and sterilize for later use.
3. Rhizobium culture medium
Glucose 10g dipotassium hydrogen phosphate 0.5g calcium carbonate 3g magnesium sulfate 0.2g yeast powder 0.4g agar 20g water 1000ml 1% crystal violet solution 1ml.
Firstly, the agar is boiled and dissolved in water, then other components are added separately, stirred and dissolved, packaged and sterilized for later use.
4. Starch nitrate medium (Gao's No.1 medium)
Soluble starch 2.0g potassium nitrate 0. 1g dipotassium hydrogen phosphate 0.05g sodium chloride 0.05g magnesium sulfate 0.05g ferrous sulfate 0.00 1g agar 2g water 100ml.
First, put the starch in a beaker, mix it into paste with 5 ml of water, then pour in 95 ml of water, stir well and add other drugs to dissolve it. Mark the outside of the beaker, add agar when it is heated to boiling, and keep stirring. After the agar is completely dissolved, make up the water loss. Adjust the pH value to 7.2 ~ 7.4, subpackage and sterilize for later use.
5. Flour agar medium
Flour 60g agar 20g water 1000ml
Mix flour into paste with water, add water to 500ml, and cook on slow fire for 30 minutes. Take another 500ml of water, put it into agar, heat and boil until it dissolves, then mix the two solutions evenly, replenish water, adjust the pH value to 7.4, subpackage and sterilize for later use.
Because of the different raw materials, the use requirements of the culture medium are different, and the storage and preservation are also slightly different. The general culture medium is easy to be contaminated or decomposed by bacteria after being heated and absorbed moisture, so the general culture medium must be kept in a damp-proof, dark and cool place. For some media (such as tissue culture medium) that need strict sterilization, they must be stored in a refrigerator at 2~6℃ for a long time. Because the liquid culture medium is not easy to keep for a long time, it is now converted into powder.
