Antitumor effect
1, The ethanol extract of Coicis Semen was injected intraperitoneally into tumor-bearing mice, which could inhibit the proliferation of Ehrlich ascites carcinoma (ECA) cells and significantly prolong the survival time of animals. Two components further separated from the extract, one can cause the protoplasm degeneration of cancer cells, and the other can stop the mitotic phase of the nucleus in the mesophase.
2. The acetone extract of Coicis Semen can also inhibit the growth of ECA by intraperitoneal injection in mice. This anti-tumor activity can be transferred to the acidic part soluble in petroleum ether. If it is injected subcutaneously, the ascites will become transparent and the tumor cells will almost disappear, but the toxicity of this part is correspondingly high. The acetone extract of Coix lachryma-jobi also has obvious inhibitory effect on solid tumor of cervical cancer 14(U 14) and ascites liver cancer (HCA).
3. The clinical application of Coicis Semen decoction can prolong the life of patients with advanced cancer, and it is found that after intraperitoneal injection of Coicis Semen acetone extract into cancer patients, the protoplasm of cancer cells has obviously degenerated through ascites examination. It was also found that the 50% ethanol extract of coix seed could promote the keratinization of cultured squamous epithelial cancer cells.
Immune function
1, neutral polysaccharide dextran mixture and acidic polysaccharide Ⅱa- 1, 2,3 and Ⅱb all showed anti-complement activity.
2. The mixture of neutral polysaccharide and glucan is highly heterogeneous. Seven glucans can be separated by HPLC, which are 1→4-linked a-D- glucans with glucosyl side chains in the 0-6 position of the main chain. Each purified glucan has different maximum absorption in iodine reaction (530nm or >: 550nm),> 500nm- glucan is more effective than 530nm- glucan, which is considered to have considerable anti-complement activity.
3. The acid polysaccharide Ⅱ a- 1, 2,3 and Ⅱb were further separated and purified to obtain anti-complement polysaccharides CA- 1 and CA-2, which contained 8- 1 1% protein in their molecules, with molecular weights of 160000 and 700000 respectively. At low dose, CA-2 showed stronger anti-complement activity than CA- 1.
Hypoglycemic, calcium and blood pressure effects
Injecting the ether extract of Coix lachryma-jobi seed (Coix lachryma-jobi seed oil) under rabbit skin can reduce the blood sugar value and blood calcium value. The water extract of Coix lachryma-jobi seed can significantly reduce the blood sugar concentration by intraperitoneal injection in mice, and three effective components, namely Coix lachryma-jobi polysaccharide A, B and C, are separated from it. These components can cause hypoglycemia in normal mice and hyperglycemic mice induced by Alloxan. Intravenous injection of coix lachryma-jobi can cause a temporary decrease in blood pressure in rabbits.
Inhibition of trypsin action
A heat-stable trypsin inhibitor was found from the bran of Coix lachryma-jobi seeds in soft shell. It is a simple protein with a molecular weight of about 12000 and contains many cysteine or cystine residues. Its molecular ratio to bovine trypsin is1:2, which shows that it is doubleheaded, and its activity is stable at pH3- 1 1 and high temperature 100℃, but the degree of thermal stability of its inhibitory activity mainly depends on the kind of solvent.
Ovulation induction
Patients with intractable anovulation can significantly improve the function of hypothalamus after taking the prescription with coix seed as the main medicine. The extract of Coix lachryma-jobi also induces ovulation in golden hamsters, and the active substances that promote ovulation are feruloyl stigmasterol and feruloyl brassinolide. 9: 1 trans-feruloyl stigmasterol and trans-feruloyl brassinolide (1) showed ovulation induction in golden hamster at a dose of 200ug/ day.
