3 Overview

Chuanxinlian Pian is a Chinese patent medicine, which is a tablet made of Andrographis paniculata by processing [1]. It has the efficacy of clearing heat and removing toxins, cooling blood and subduing swelling. It is used in the treatment of evil toxicity, cold and fever, sore throat, mouth and tongue, cough and cough, diarrhea and dysentery, fever and astringent pain, carbuncle and swollen sores, and poisonous snake bites.

4 The efficacy and main treatment

The penetrating lotus tablets have the effect of clearing away heat and detoxification, cooling the blood and eliminating swelling. It can be used in the treatment of evil toxicity, cold and fever, sore throat, mouth and tongue, cough and cough, diarrhea and dysentery, fever and astringent pain, carbuncle and sores, snake bites.

5.3 Preparation

Take Chuanxinlian Pian and extract it twice by hot immersion in 85% ethanol for two hours each time, then merge the extracts and filter them, then recover the ethanol in filtrate, concentrate them to the right amount, dry them, add appropriate amount of auxiliaries and make granules. Add the appropriate amount of auxiliary materials, made of granules, dry, pressed into 1000 tablets (small tablets) or 500 tablets (large tablets), sugar coated or film coated, that is obtained.

5.4 Character

This product is sugar-coated or film-coated tablets, gray-brown to brown after removing the coating; bitter taste.

5.5 Identification

Take the filtrate under [Content Determination] as the test solution. Take 0.5g of Andrographis paniculata control herb, add 30ml of methanol, ultrasonic treatment for 30 minutes, filtration, the filtrate was concentrated to about 5ml, as the control herb solution. Then take the dehydrated Andrographis paniculata lactone control, add methanol to make a solution containing 1mg per 1ml, as the control solution. According to thin-layer chromatography (2010 Pharmacopoeia, Appendix VI B), absorb 5μl of each of the above three solutions, respectively, on the same silica gel GF254 thin-layer plate, trichloromethane - ethyl acetate - methanol (20:15:2) as an unfolding agent, unfolded at 28 ℃, remove, and let dry. Under the ultraviolet light (254nm), in the chromatogram of the test article, in the corresponding position with the chromatogram of the control herb and the control article, the spots of the same color were shown; sprayed with 2% 3,5 dinitrobenzoic acid ethanol solution and 2mol/L potassium hydroxide solution in equal volume of the mixture (prepared for clinical use), and then immediately examined under the sunlight. In the chromatogram of the test product, in the corresponding position with the chromatogram of the control herb and the chromatogram of the control product, the spot of the same color.

5.6 Inspection

5.7 Determination of content

Determined by high performance liquid chromatography (2010 Pharmacopoeia, Appendix VI D).

5.7.1 Chromatographic conditions and system suitability test

Octadecylsilane-bonded silica gel was used as filler; methanol-water (60:40) was used as the mobile phase; the detection wavelength was 254 nm; the theoretical plate number should not be less than 2,000 based on the peaks of the dehydrated andrographis paniculata lactone.

5.7.2 Preparation of the control solution

An appropriate amount of the control product of dehydrated andrographis paniculata lactone was taken.

Take the appropriate amount of dehydrated Andrographolide control product, precision weighing, add methanol to make a solution containing 0.1mg per 1ml, that is obtained.

5.7.3 Preparation of test solution

Take 20 tablets (small tablets) or 10 tablets (large tablets), remove the coating, precision weighing, research fine, take 0.5g, precision weighing, placed in a stoppered conical flask, precision add 25ml of methanol, tightly stoppered, weighed, immersed in 1 hour, ultrasonic treatment (power 250W, frequency 33kHz) for 30 minutes, cooled, and then weighed. Make up the lost weight with methanol, shake well, filtered, precision measure 10ml of filtrate (the remaining filtrate for backup), added to the neutral alumina column (200-300 mesh, 5g, inner diameter of 1.5cm), eluted with 20ml of methanol, collect the eluent, placed in a 50ml measuring flask, add methanol to the scale, shaking, that is, obtained.

5.7.4 Determination

Precisely aspirate 10μl each of the control solution and test solution separately, inject into the liquid chromatograph, determination, that is, obtained.

Each tablet of this product contains dehydrated Andrographis paniculata lactone (C20H28O4), the small tablets shall not be less than 4.0mg, the large tablets shall not be less than 8.0mg.
