Pharmacological Actions
1. Antibacterial Actions Qu Shuyan reported that the ethanol extract of Portulaca oleracea had a significant inhibitory effect on Shigella and Fusarium. Water decoction has inhibitory effect on Shigella, Sonnen, S. and F. dysenteriae, but dysentery bacilli can produce significant resistance after several passages in Amaranthus broth. Alcoholic extract or decoction also has inhibitory effect on Escherichia coli, S. typhi and Staphylococcus aureus; it also has different inhibitory effect on some pathogenic fungi such as S. aureus. It has no inhibitory effect on Mycobacterium tuberculosis; it is ineffective in mice infected with Escherichia coli by intraperitoneal injection with alcoholic extract or water decoction of residue after alcoholic extraction.In 1992, Ma Muying reported the bacteriostatic effect of wild amaranth sold in the vegetable market of Shenzhen City on a variety of common food-contaminating bacteria. The results showed that amaranth had strong inhibitory effects on a variety of common food contaminating bacteria. Even diluted to a concentration of 12.5%, it still had a certain inhibitory effect. Especially, it has strong inhibitory effect on many bacteria, such as Escherichia coli, Salmonella, Proteus, Shigella, Staphylococcus aureus, Bacillus subtilis, Bacillus cereus, etc., while it does not inhibit Garcinia cambogia, and its inhibitory effect on Mycobacterium smegmatis is not obvious. 2.Effect on smooth muscle In 1985, Qu Shuyan reported that the extract of Amaranthus tinctorius (made by decocting and concentrating in water and adding alcohol to remove the precipitate) and its isolated crystalline potassium chloride have obvious excitatory effect on guinea-pig, rat and rabbit in vitro, and on the in situ uterus of dog. 2 ml of Amaranthus injection (equivalent to 5-10 g of the raw drug) was stronger than with ergometrine 0.2 mg, and weaker than with posterior pituitary hormone 10 units; whereas with 4-6 ml it was similar to posterior pituitary hormone 10 units. Potassium chloride crystals isolated from Amaranthus tinctorius had a significant contraction effect on isolated ileum of rats, and the strength of its action was similar to that of prostaglandin E1, 200ug, and neostigmine 0.25mg, while 20% decoction 0.2ml had an inhibitory effect on the small intestine of isolated guinea-pigs. After that, foreign scholars SunbhanichM et al. studied the effects of aqueous juice and boiling water extract of freshly squeezed amaranth plants on intestinal peristalsis of isolated guinea pigs and contraction of isolated atria and trachea. The results showed that for isolated ileum, both the aqueous juice and the boiling water extract resulted in increased tension, amplitude, and frequency of contractions. This effect was similar to that of acetylcholine and was dose-dependent. However, the increase in contractile tension and peristalsis could be slightly blocked by atropine. Both types of preparations showed dose-dependent effects on cardiac myocardial contractility and rate of contraction, as well as relaxation of isolated tracheal strips. These effects were similar to those produced by isoprenaline and were completely blocked by cardiac glycosides (2.1x10(-5) mol/L). The results of the action of Mastodon on isolated guinea pig ileum and atria suggest that the drug may act as a relaxant and cardiotonic agent, respectively, and that its bronchodilatory action on bronchial tubes may be used in the treatment of bronchial asthma.
3. Action on skeletal muscle:
OkwuasabaF et al. 1987 reported that the aqueous extract of Amaranthus tinctorius had unique properties of diastolic in isolated and in vivo skeletal muscle, and that the aqueous extract was effective when used topically in skeletal muscle ankylosis due to spinal cord injury. The authors further investigated the pharmacological activities of various extracts of Amaranthus and compared them with the aqueous extract. The results showed that the effects of the various extracts of Amaranthus (methanol (MEE), ether (DEE), and dialyzable (DIF) extracts) were similar to those of the aqueous extract (AEE) in terms of their inhibitory effects on the phrenic nerve of the rat and the lateral diaphragm (MS and NS) and on the rectus abdominis muscle of frog specimens.
All three extracts:
3.1. produce a biphasic response consisting of an initial increase in twitch tension followed by a prolonged and sustained inhibition;
3.2. significantly reduce the ratio of twitch/tonicity in contractures induced by K+ and caffeine;
3.3. attenuate contractures of rectus abdominis muscles induced by the agonist nicotinic acid. The non-dialyzable (NDF) extractor showed very slight muscle relaxation without attenuating agonist nicotine-induced contractures. The mechanism of action can be hypothesized to be as follows: The aqueous and organic solvent extracts of Amaranthus produce muscle diastole by interfering with a variety of calcium pools associated with excitation-contraction couplings such as initiating calcium and intracellular calcium pools. Caffeine-induced contraction occurs in part through mobilization of sarcoplasmic reticulum (SR) calcium pools, and interference with the mobilization of such pools partially explains the observed attenuation of caffeine contraction by various different amaranth extracts. Other effects of these extracts such as inhibition of convulsions (MS) and tonic tension can be explained by a similar mechanism. The order of strength of IC50 of these extracts was DIF ≥ MEE>DEE. since methanol has a lower partition coefficient than ether, it is possible that methanol extracts contain more of the active ingredient for the muscle-diastolic effect of Amaranthus. Based on the significant effect of DIF and the absence of skeletal muscle diastolic effect of the 100% acetonitrile fraction previously extracted using high-pressure liquid chromatography, it can be assumed that the active ingredient of Amaranthus equinus may include polar components.
4. Effects on uterus: Li Shourui reported in 1979 that Amaranthus had two opposite effects on the uterus of animals, one was excitatory, which was the potassium chloride fraction in Amaranthus, and the other was inhibitory, which was the organic component in Amaranthus.
