1, mother seed production
1) Add potatoes into the leaching solution of termite nests, heat and boil, and filter;
2) adding water into the cooking liquid to make it reach 1000ml, adding watercress powder, boiling until it is completely dissolved, and then autoclaving;
3) Put the sterilized culture medium on a sterile operating table and pour it into a Petri dish or other culture container while it is hot;
4) Inoculating termitomycetes after cooling.
2. Production of bacterial bags
1) Weigh the ingredients respectively, then mix them evenly and pile them up; ?
2) After 2 hours, check the water content, grasp the culture material by hand, and there are water drops between fingers, but the water drops are not enough; ?
3) culture bag 17cm? ×35cm or 15cm×35cm polypropylene bag material, two sets of collars, and no cotton plastic cover to seal the bag mouth;
4) Sterilize according to the general procedure, and then inoculate the strains of termitomycetes in the sterile room.
Step 3 harvest
When the cap of the fruiting body of the termites is about to become straight without cracking, it can be harvested. When harvesting, you can gently grasp the base of the seedling stem, cut it off along the lower edge of the expanded stem with a sharp knife and pull it up. Can be harvested for 3-4 times, and the biotransformation rate is 50%-60%.
Fungal culture:
Put the inoculation bag into a clean culture room, control the bag temperature at 20 ~ 25℃, and keep the room dark for fungus culture. Generally, hyphae can grow all over the bag in 40 ~ 60 days, and when there are spikes or coral-like tumor spots the size of rice grains on the bag wall, they can be moved into the soil.
Seasonal arrangement:
When the temperature is stable at 12 ~ 24℃, bag cultivation can be arranged. After 40 ~ 60 days of cultivation, mycelium will grow in the bag and then be planted in the soil. At this time, the temperature has risen above 25℃, which is the best season (summer) to promote germination and fruiting.
