2, tank sterilization. Put the culture medium into a jar and seal it as required. A small fermentor (5 liters) can put the jar into a large sterilizer for sterilization (1 15 degrees Celsius for 30 minutes). Steam sterilization in large fermenters (10L and above) (120℃ for 30 minutes).
3. After the tank is cooled, put it on the fermentation platform and install it well. Turn on the cooling water, turn on the power supply of the air pump, connect the vent pipe to start ventilation, adjust the intake knob to make the ventilation appropriate, keep the tank pressure at 0.05MPa, and set the temperature, pH, stirring speed, etc. The slope of the dissolved oxygen electrode is calibrated at a certain speed and ventilation, and the value is 100%.
4. When the temperature is stable and all parameters are correct, inoculate pre-shaken seeds, start fermentation timing, and record all parameters.
5. After the fermentation, clean the tank and the electrode, insert the pH electrode into the 3M triangular bottle for later use, and cover the probe of the dissolved oxygen electrode with a protective sleeve for later use.
